@Hulskof, ik vraag me af of deze journalist soms ingehuurd is voor verspreiding van enig optimisme. De beurs lijkt er tenminste positief op te reageren. De indruk wordt gewekt dat de AFM13 data goed zijn (Paukenschlag). Ik had ook graag meer diepgang willen hebben over de reorganisatie en het vertrek van Hoess en nog 2 grootverdieners. Hierover wordt in in alle talen gezwegen, nee dit is geen onderzoeksjournalist.
Mee eens. Het stuk brengt geen verheldering en stelt alleen wat al bekend was. Zoals eigenlijk vrijwel alle stukken, aangezien bedrijven zelden of nooit openheid van zaken geven. Als particuliere belegger moet je altijd maar gissen wat er achter bepaalde beslissingen schuilgaat en ik denk wel eens dat ook institutionele beleggers het fijne niet weten van wat er speelt. Enfin, nog enkele weken geduld, dan weten we (veel) meer.

NKX101 van Nkarta dat NK cellen contact laat maken met NKG2D receptoren in geval van AML is vandaag op een spaarvlammetje gezet na eerdere positieve berichten:www.onclive.com/view/nkx101-demonstra... www.investors.com/news/technology/nka... Weer een tegenvaller voor de NK sector. De koers is dan ook 29% lager. Gelukkig is Daina Graybosch niet echt negatief geworden tav NK cellen: "Bears will inevitably argue Nkarta's decision suggests the cells it was using — known as natural killer cells — aren't potent enough for any use, Leerink Partners analyst Daina Graybosch said in a report. But she disagrees with that sentiment".

@Hulskof. Je moet ergens beginnen :-). De berichten aangaande Nkarta werken natuurlijk niet in het voordeel van de NK sector. De één na de ander stapt over op autoimmuunziekten zo lijkt het. Ik begrijp niet zo goed waarom het bij Nkarta steeds mis lijkt te gaan na initieel juichende fase 1 trials. De hamvraag is nu of de NK engagers van Affimed zoveel beter zijn dan de CAR-NK oplossingen van Nkarta. Ik denk dat het door mij aangehaalde citaat van Diana Graybosch mij enigszins geruststelt. Graybosch heeft tenminste een gedegen biomedische achtergrond. Ooit heeft Takeda een licentie genomen op de CD19 CAR NK activiteiten van Anderson. Heb jij daar ooit nog iets over gehoord? Ik verwacht dat de Affimed oplossing superieur is aan CAR-NK. Hoezo doorbraak? Alles was knip en klaar (trials opgezet en lopende), maar vanuit de markt kwam het signaal dat er iets moest veranderen (lage koers die niet wilde oplopen ondanks de ingeslagen weg) waardoor er door emissies geen geld meer kon worden opgehaald en er per se een partnership gesloten moest worden. Dat is Hoess om wat voor reden ook niet gelukt. Hij had weinig om mee te strijden, doordat de succestrials met Anderson niet werden voortgezet. Het is nu gewoon wachten op een herhaling vd Anderson data (of hopelijk zelfs een verbetering daarvan). Wordt dat een mislukking, dan is het einde verhaal.

We weten dat Anderson inmiddels de opzet (shift van verse cellen naar diepvries) van deze studie ingrijpend heeft aangepast. Dus gaat het naar analogie van Affimed nog veel langer duren? Ik vind dat de FDA trouwens op dit gebied overdreven kritisch is: bij NK cellen hoef je alleen maar te letten of er niet te veel T cellen tussen zitten? Takeda kan zulks beter opvangen dan een minispeler als Affimed. Maar goed te lezen dat Takeda hier nog steeds geld in stopt.

Nu we toch in de wachtkamer zitten is het wel interessant andere CD30/Hodgkin therapieën te bestuderen. CD 30 antigen escape lijkt niet veel voor te komen bij CAR-t onderzoek dat een aantal jaren geleden is uitgevoerd door een aantal universiteiten in de VS. De ORR en CR percentages zijn inferieur aan die van AFM13+NK cellen. Het bijwerkingen profiel schijnt oke te zijn en ook met de durability zit het het wel goed. Hodgkin blijkt een kanker met een vijandige omgeving voor therapieën (tumor micro environment). Toch beter geschikt voor NK cellen? Ik had ergens gelezen dat:www.horizonscangeneesmiddelen.nl/gene... odronextemab ook een CD30 antagonist was, maar die zat er dus gelukkig naast. Het blijkt om een CD3/CD20 bi specifiek antilichaaml te gaan tegen non-hodgkin. Voor dit middel is door de FDA een complete response letter afgegeven.

Program Updates AFM24 (EGFR/CD16A tumors) As of January 4, clinical response update to the Phase 1/2a AFM24-102 trial in EGFR-wt NSCLC reported 4 confirmed responses, including 1 CR, 3 PRs, and 7 stable diseases in 15 heavily pre-treated patients, resulting in a disease control rate of 73 percent. All patients were pre-treated with platinum-based chemotherapy and checkpoint inhibitors PD [L]-1. Based on the promising results, Affimed expanded enrollment in this cohort to 40 patients. In addition, the company continues to enroll in the EGFR-mut NSCLC cohort for a planned number of 25 patients. Mature PFS data from the first 15 patients from the EGFR-wt cohort and response data from the EGFR-mut cohort are expected in Q2 2024. Acimtamig (AFM13; CD30/CD16A tumors) Initial safety and efficacy data from the LuminICE-203 (AFM13-203) study expected in Q2 2024. LuminICE-203 is a Phase 2 clinical study investigating acimtamig in combination with Artiva’s AlloNK cells in patients with r/r classical Hodgkin lymphoma (HL). Safety Review Committee meeting and initiation of enrollment in cohorts 3 and 4 is expected mid-April. Updated Phase 1/2 AFM13-104 study data demonstrated an ORR of 97% and a CR rate of 78% in 32 patients with r/r HL, presented at ASH 2023. As of the cutoff date, the median EFS was 9.8 months with 84% patients alive at 12 months. The median duration of response was 8.8 months. The treatment regimen continues to demonstrate a good safety and tolerability profile with no cases of cytokine release syndrome (CRS), immune effector cell-associated neurotoxicity syndrome (ICANS) or graft versus host disease (GVHD) of any grade. The oral presentation included a total of 42 patients enrolled, with 36 patients treated at the recommended Phase 2 dose (RP2D). All patients were heavily pretreated and refractory to their most recent line of therapy with active progressive disease at the time of enrollment. AFM28 (CD123/CD16A) Completed enrollment of the fifth cohort, recruiting patients in the sixth and final cohort in the multi-center Phase 1 open-label, dose-escalation study (AFM28-101), of AFM28 monotherapy in CD123-positive r/r AML. No dose-limiting toxicities were reported. Further clinical development of AFM28 is planned in combination with an allogeneic off-the-shelf NK cell product.
Corporate Restructuring: The corporate restructuring announced earlier this year has been fully implemented. The restructuring included a streamlining of operations resulting in a 50% reduction in the Company’s workforce. The Company is focusing resources on advancing its clinical programs, AFM24, acimtamig, and AFM28, through the various stages of development. Upcoming Milestones: Data from the NSCLC expansion cohorts in the Phase 1/2a AFM24+atezolizumab combination study expected in Q2 2024. Brian heeft the call beluisterd en is enthousiast. Dr Harstrick is sounding very confident/optimistic that PFS will come through for AFM24-102. They will have a detailed plan to proceed with AFM28 in combination with allogeneic NKs! The only possible option that I can foresee is a hookup with an interested party. $$ I need to listen to the call again but don’t think they will be at ASCO. Only 4 analysts on the call, funny how sell-side drops off when they perceive there’s no fees to be made. A definite positive for investors. Phase 2 LuminICE: “Safety Review Committee meeting & initiation of enrollment in cohorts 3 & 4 is expected in April.” Me: Judging by their use of the compound sentence (above) initial safety in cohorts 1 & 2 is a given. Will be getting the eagerly awaited initial efficacy in Q2. For cohorts 1 & 2 (lower dose). Starting 6th and final cohort in AFM28 Phase 1 dose escalation. No dose limiting toxicities thus far. @Hulskof, bedankt voor je bericht van Brian. Wie weet zijn ze (met een partner ) de eersten die een effectief middel in de strijd tegen AML hebben. Dat is alleen al miljarden waard, tenminste getuige de bedragen die bij Argenx betaald zijn. De beurs is vandaag minder uitbundig, ik hoop zo dat Brian (en ik ) gelijk krijgen. Het is ook fijn te vernenmen dat alles gewoon door marcheert ondanks dat Hoess en die CFO thuis zitten.
