Graag gedaan en dank voor de link. Heb al geruime tijd met veel waardering de bijdragen van o.a. Prof. Dollar en Ivet gelezen. Derhalve blij dat ik ook een keer een zinnige bijdrage kan leveren aan dit kwalitatief/inhoudelijk goede forum.
Dit was inderdaad de missing link die nog ontbrak in mijn verhaal. Hoewel ik het ook niet zeker weet, lijkt het mij alleszins redelijk om te veronderstellen dat de rechten op de vector zijn mee verhuisd van AMT naar UniQure.
In mei 2014 zou de hemofilie studie volgens clinicaltrials afgerond moeten zijn ('primary completion'). Het zou mooi zijn als dan 180 dagen na verschijnen prospektus t.b.v. IPO mooie resultaten gebracht kunnen worden. Wellicht samen met clinical data met betrekking tot porfyrie, die in 2014 worden verwacht en start clinical trial San Fillipo. De porfyrie trial is overigens niet aangemeld in clinicaltrials.gov, waarschijnlijk omdat het een studie betreft die in Europa wordt uitgevoerd. Maar dat is niet iets voor op het hemofilie draadje...
Dat zou een strategie/ het vooruitzicht van de beursgang kunnen zijn, na de locked up positieve resultaten hemo b. Goed dit te lezen !

Het volgende citaat is gekopieerd uit het stuk dat Bilbo3 op 7 november in dit draadje heeft gepost: “I think it’s incredibly exciting, and I say that even though these people are my competitors,” she said. Dr. High is listed as a co-author of the report because her laboratory helped monitor the patients and provided proof for regulators that the virus would not insert its human gene into the patients’ sperm and make the change hereditary. Ze is co-auteur omdat haar lab voor deze publicatie in de NEJM heeft aangetoond dat het ingebrachte gen niet bij voortplanting kan worden doorgegeven (wat tot zorgen van de toezichthouders had kunnen leiden als dat wel zo was). Dit zou ze theoretisch op verzoek van iedere vector voor iedere willekeurige genetische aandoening kunnen testen, vermoed ik. Het staat mijns inziens dus los van welke vector wordt gebruikt voor het inbrengen van het gen en hoe effectief dit gen is als gaat om het tot expressie brengen van factor IX, dat is niet wat door de 'Philadelphia' groep is bestudeerd. Dat wordt door de St. Jude's collaboration onderzocht en dat is wat voor ons als beleggers in UniQure relevant is, aangezien de vector door UniQure geleverd wordt, en ik veronderstel straks ook het eindproduct. Wellicht heeft dr. High ook een bijdrage geleverd aan het meten van de immuunrespons van het patienten die het medicijn kregen toegediend, volgens mij haar specifieke deskundigheid. Het levert haar misschien geen commerciele rechten op, maar wel een co-auteurschap in een van de meest gerenommeerde medische tijdschriften (NEJM). Maar misschien is iemand in staat om a.s. maandag deze vraag, of UniQure en Spark inderdaad elkaars competitors (aldus dr. High) zijn, aan Aldag zelf te stellen. Dan weten we het zeker, ik interpreteer alleen maar. Aldag die zelf trouwens ook voor een (niet onaanzienlijk) deel in certificaten B wordt uitbetaald, en dus voor een groot deel dezelfde belangen als de overige certificaat B-houders heeft.

ik gok dat Frenky T. hierop doelde als antwoord op jouw vraag in een eerdere posting: "Dit was inderdaad de missing link die nog ontbrak in mijn verhaal. De rechten op de vector, de patenten...voordat ik in UniQure ging uitbreiden heb ik mij ervan vergewist dat intellectueel eigendom ook mee was overgegaan van AMT naar UniQure, want zonder dat is het een lege huls. Het schijnt een verdienste van Aldag te zijn dat die rechten behouden zijn voor UniQure. Het is ook een goede zaak dat A,B en C-aandelen gelijkgesteld worden...derhalve denk ik wel, dat mócht er overnamebelangstelling zijn, dat het vóór de IPO strategisch het gunstigst zal zijn voor de overnemer, daar ze m.i. alleen de A-aandeelhouders over de streep hoeven te trekken. Ik weet niet of daar op dit moment afspraken over zijn gemaakt in aanloop naar de IPO. ivet

Wat ik bedoelde is dat Aldag heeft gezegd dat ze hun (dus UniQure) technologie hebben benut en dat is allemaal beschermd. Ze kennen Spark en zijn volledig op de hoogte. Moet ik uit 1. en 2. concluderen dat het eigenlijk om 2 studies gaat? Een fase I/II studie in samenwerking met St. Jude Children’s Research Hospital; waarbij een AAV8 vector wordt gebruikt (Dose-Escalation Study Of A Self Complementary Adeno-Associated Viral Vector For Gene Transfer in Hemophilia B) Deze studie is in december 2013 nog geupdate/verified in clinical trials.gov en recruteert nog steeds (..) proefpersonen (http://clinicaltrials.gov/ct2/show/NCT00979238?term=hemophilia+B&rank=16). Volgens bovenstaande persberichten liggen de commerciele rechten voor het eindproduct evident bij uniQure (AMT). En daarnaast een studie, ook fase I/II, van uniQure zelf met een AAV5 vector (die een sterke affiniteit voor levercellen, waar je hemofilie geneest, heeft). Die studie moet nog van start gaan (2e helft 2014, in samenwerking met, en voor de helft gefinancierd door Chiesi).

www.ehc.eu/fileadmin/EHC-conference/2... Van maart 2010 tot oktober 2013 zijn er dus 10 patienten geincludeerd in de studie. 10 van de 18 die nodig zijn voor de fase I/II trial. Als je dit inclusietempo extrapoleert zijn er dus nog 2 jaar nodig om aan de 18 te komen.
Desalniettemin zien de getoonde resultaten er fantastisch uit. Met ook weinig/geen safety concerns.

Het vinden van geschikte en bereidwillige patienten voor inclusie kost immers veruit de meeste tijd (toedienen van therapie is kwestie van uren of korter, het monitoren van acute bijwerkingen kwestie van een dag, en het in het bloed vervolgen van de voordelige/nadelige effecten van het medicijn een kwestie van weken/maanden voor de fase I/II). En het aantal van circa 12 is kennelijk 13 tot 16 patienten geworden? De St. Jude's studie met
AAV8 -vector (http://www.ehc.eu/fileadmin/EHC-conference/2013/Gene_therapi_Nathawani.pdf) heeft nu al meer tijd nodig om uberhaupt aan het beoogde aantal patienten van 18 te komen, zo lijkt het.
In zekere zin loopt St. Jude voorop. De belangstelling gaat daarbij niet uit naar de verpakking (AAVx) maar naar de inhoud ervan (het gen hFIX). uniQure gebruikt namelijk hetzelfde gen: "St. Jude is currently conducting a Phase I/II, open label, dose escalation clinical trial in this indication with a gene therapy consisting of an AAV8 vector carrying the same therapeutic hFIX gene that we are using in AMT-060." uniQure zegt ook iets over de commerciële rechten. Maar hoe die zin gelezen moet worden is mij niet helemaal duidelijk: "AMT-060 consists of an AAV5 vector carrying a therapeutic hFIX gene cassette that we have exclusively licensed from St. Jude." Heeft uniQure nu alle commerciële rechten op de "hFIX gene cassette"? Of enkel in combinatie met AAV5? Het is mij niet duidelijk. Daarnaast is er ook een discussie gaande over het alleenrecht op AAV5 in bredere zin. uniQure heeft wel aan er zelf eigenschappen aan toe te voegen: "In collaboration with Chiesi, we are developing AMT-060 as a gene therapy consisting of an AAV5 vector carrying an hFIX gene that we have exclusively licensed from St. Jude, in which we have altered the codons to maximize expression, together with the insertion of a liver-specific promoter, LP1. We produce this vector with our insect cell-based manufacturing process." De studie kenmerken: "We are currently planning to initiate a Phase I/II clinical trial of our AMT-060 product candidate in the second half of 2014 under our co-development agreement with Chiesi. We intend to file an IND and IMPD in order to initiate clinical development of AMT-060 and anticipate that the trial sites will be in Europe. We are designing this clinical trial as a multicenter, open label, prospective, interventional, single dose, dose escalation study to investigate the safety and tolerability of AMT-060 in patients with severe hemophilia B. We currently expect to recruit 13 to 16 male patients between the ages of 18 and 35 with severe, genetically confirmed hemophilia B. The primary objective of this clinical trial will be to assess the safety of systemic administration and determine the maximum tolerated doses."
"AMT-060 consists of an AAV5 vector carrying a therapeutic hFIX gene cassette that we have exclusively licensed from St. Jude." Alleen de hFix gene cassette is exclusief in licentie genomen. Alleen de hFix gene cassette is exclusief in licentie genomen. Daar heeft Uniqure een andere vector (AAV5 ipv AAV8) aan toegevoegd. Dus de eigen vector en het ingelicentieerde gen is samen AMT-060.
Zo las ik het ook... echter, Spark Therapeutics zegt zich ook te baseren op eerder onderzoek... waaronder dat van St. Jude. Willen zij hetzelfde gen gebruiken? Of hebben ze een variant bedacht om die licentie te 'ontwijken'?
