Welke firmanaam komt straks op het pillenpotje in EU. Het zou uiteraard Gilead worden, maar met de nieuwe commerciele structuur gaat de handelsvergunning naar Gala maar blijven de rechten bij Gilead.
Dus kan Gala een pillenpotje met Gala op het etiket gaan verkopen of prijkt de naam Gilead op het stickertje?

Maar is dat reden om het aandeel te kopen? Niet als sprake blijkt te zijn van fraude. Gisteren daalde de koers met een lichte 1,2% op 78,18 euro. De afgelopen week is het aandeel met 20% gedaald doordat de Amerikaanse toezichthouder te veel risico op ernstige bijwerkingen ziet voor het reumamedicijn filgotinib. Galapagos en de Amerikaanse partner Gilead hebben de poging gestaakt om het medicijn alsnog goedgekeurd te krijgen in de VS. De samenwerking tussen beide partners is gewijzigd. Tegenover een wegvallende inkomstenstroom aan royalties is er voor Galapagos meer ruimte ontstaan om zelfstandig actief te zijn op de Europese markt. De koersgeschiedenis van 2018 en 2019 lijkt zich te herhalen. In beide jaren werd een bodem gevormd rond de 75 euro in het begin van het jaar. In het voordeel van koopjesjagers is de lage stand van de RSI en het prijsmomentum dat op zoek is naar een bodem. Fundamenteel staat daar een negatieve koers/winst tegenover. Het aandeel hunkert naar een nieuwe positieve impuls. Het vertrouwen van de beleggers in het aandeel is echter beschaamd en zwaar op de proef gesteld. Dat maakt de start van een nieuwe langdurige opgaande beweging er niet gemakkelijker op. Inmiddels is het Amerikaanse advocatenkantoor Pomerantz LLP een onderzoek gestart naar aandelenfraude of andere juridisch verhaalbare praktijken bij Galapagos. Pomerantz heeft meer dan 80 jaar ervaring in dit soort van juridische gevechten.
Er zijn zeker momenten geweest dat mt wist dat US er niet rooskleurig uit zag en dat plaatje ons zeker niet heeft voorgeschoteld. In hoeverre daar een rechter in mee gaat kan ik niet beoordelen Maandag stap ik uit met meer dan 50% verlies. Ik zie betere kansen elders om dat terug te verdienen. Loop al tijdje mee , maar heb me toch teveel laten verleiden door een veel te enthousiaste ceo.

Zoals ik het begrepen heb, is het verschil in werking tussen 100mg en 200mg te klein. Zelfde bij Abbvie's Rinvoq. Die hebben 30mg niet gefiled, enkel 15mg. Maar voor UC gaan ze nu wel 45mg filen. Vermits de mindere dosis niet werkt, zal de kans op goedkeuring er wel zijn.
Als 30mg te veel bijwerkingen heeft voor reuma, hoe kan 45mg dan aanvaardbaar zijn voor? Filgotinib onderscheidde zich toch met het veiligheidsprofiel?

Ik zag Takeda vroeger wel als potentiële overnemer van Morphosys, maar daar is het niet van gekomen. Op de huidige koers van Galapagos is het risico op een bod van een derde partij helaas weer toegenomen. Het liefste zie ik nog steeds een zelfstandig Galapagos.
Het is toch moeilijk om met de volgende valuation vrede te nemen. -Filgo RA => €18/share (success rate / launch: 0% US 2021 + 100% EU 2021 + 100% JP2022) -Filgo UC => €5/share (success rate / launch: 90% US 2022 + EU 2021 + JP 2022) -Filgo CD => €3/share (success rate / launch: 57% US 2023 + EU 2023 + JP 2023) -Ziritaxestat IPF => €15/share (success rate / launch: 42% Global 2023) -GLPG12025 IPF => €2/share (success rate / launch: 23% Global 2026) -Ziritaxestat SSC => €2/share (success rate / launch: 21% Global 2024) -Net Cash => €4.7Mio => €72/share -Overhead => -€1.7Mio => -€27/share Total Valuation: €90/share Maar nu zou ik hier reeds "blij" mee zijn gezien de koers maar blijft dalen. Om zelf op mijn vraag te antwoorden,- ik zie nog "toekomst" in Gala en ga voor "LONG". Kan me niet van de indruk ontdoen dat iedereen bij Gala toch ergens uit "onvrede" met de FDA beslissing een soort van "eergevoel" heeft om terug naar de hogere regionen te (over)stijgen. Daarmee bedoel ik een koers welke binnen de 2j terug op "niveau" staat..+/-€150 (en hopelijk hoger mits wat meevallers / overnames). Ook met de cashburn en partnership Gilead zal er naar een hogere versnelling geschakeld moeten worden om investeerders terug aan boord te krijgen / niet te verliezen. In tussentijd kruip ik onder een steen,- en probeer ik mezelf wijs te maken dat "alles" altijd (deels) goedkomt. Momenteel een megaverlies van 60% (virtueel) en een veel te grote positie in Gala. Plan is om mijn "geloof" proberen aan te houden in Gala en verliezen iets in te perken in 2021 en te blijven "vertrouwen" in Gala. Mijn "geluk" zoeken ergens anders vind ik persoonlijk ook riskant na deze enorme tegenvaller.

Wat zouden ze willen overnemen? Iets commercieels dat ze via hetzelfde verkoopapparaat kunnen verkopen? Versterking van de pijplijn? Ik zou liever extra omzet dan extra cashburn zien.

Filgotinib ( www.glpg.com/filgotinib ) Gilead will not pursue Jyseleca launch in RA in the US and the Ph3 trials in psoriatic arthritis (PsA) and ankylosing spondylitis (AS) will also be discontinued. Gilead remains on board for ulcerative colitis (UC) and Crohn’s Disease (CD). Submission in Japan and positive CHMP opinion for UC are expected in H1 2021, whilst the Ph3 DIVERSITY readout in CD is anticipated for H1 2022. Further information on the IBD US filing is anticipated after consultation with the FDA, which will include discussing MANTA and MANTA-RAy results. Positive MANTA readouts will be crucial for any Jyseleca US approval. Week 26- data are expected in H1 2021. However, the FDA indicated that patients who experienced more than 50% sperm count reduction and have not recovered by week 26 will need to be followed up to 52 weeks. As a consequence, any decision on the future of Jyseleca in the US for IBD is now likely to come in 2022. Geoff Porges (Wall Street Analyst at Leerink Partners) suggests that the company won’t proceed with that indication either, given the drug’s modest showing in clinical trials in another inflammatory bowel disease, ulcerative colitis. “Gilead shared with us that they would not proceed with commercializing the drug in CD unless they see best-in-class data,” Porges said. To further support business transfer and development costs, Gilead will pay Galapagos EUR 160m to Galapagos in the course of 2021 and 2022 (€110m in 2021 and €50m in 2022). GILEAD / GALAPAGOS Now that filgotinib’s path to the US market is definitely closed in RA, it is time to take a step back and review filgotinib’s market opportunity as this new development could have a broader impact on its commercial perspectives. Even though it is believed that Gilead is still fully committed to the IBD & Crohn’s Disease market. The future will be in inflammatory bowel conditions where for now Gilead is pushing on. Rights have been handed back in RA, psoriatic arthritis, ankylosing spondylitis and non-infectious uveitis. While Gilead remains committed to the partnership and inflammation, most analysts get the sense that the company is more focused on oncology with the recent acquisitions of Immunomedics and Forty Seven. While we have been told in the past that MANTA data (assessing filgo’s testicular toxicity) was not needed to submit in the US, the understanding from last August CRL is that 26-week follow-up data was now going to be necessary. Following the Conference call, it now appears that it is very likely that FDA will ask to see up to one year recovery period after the 26-week data. The further we go, the less clear the path seems which is all the more worrisome considering that the rest of the US commercial opportunity is at stake. As US commercial opportunity fades away this new uncertainty is very unwelcome as it may be another reason for Gilead to put an end to filgotinib’s US journey. Gilead and Galapagos disclosed that the FDA now wants to see 52-week follow-up forpatients in the MANTA/MANTA-RAy studies that had a >50% decrease in semen parameters by week 26. Data for week 26 will be available by mid'21 and was supposed to sufficient for continuing the review in RA and future indications. As a consequence any decision on the future of Jyseleca in the US for IBD is now likely to come in 2022. Galapagos CMO noted that there was an un-blinded group looking at the MANTA/ MANTA-RAy data that will communicate with the EMA and regulators in Japan, where filgotinib gained approval and patients are being treated with the drug, to ensure any safety signal from the MANTA studies is relayed to these agencies. It's not clear if a safety signal is seen, if there is risk to filgotinib in these markets but this puts even more importance on these studies. Gilead has decided to retain worldwide rights outside Europe but should the toxicity concerns be confirmed, a complete exit from this product is surely inevitable. Even though Galapagos Market cap is approximately equal to its current cash position, the uncertainties remain. It is fair to doubt Gileads commitment as it has shown in the past it is capable of making ruthless decisions if needed. Some observers also question how independent Galapagos now really is, because of the deal made last year it gave Gilead two members on the board who could “potentially veto anything they don’t like”. Galapagos funds and leads all discovery and development autonomously until the end of Phase 2. After the completion of a qualifying Phase 2 study, Gilead will have the option to acquire a license to the compound outside Europe. If the option is exercised, Galapagos and Gilead will co-develop the compound and share costs equally. Gilead will maintain option rights to the programs through the 10-year term of the collaboration and for up to an additional three years thereafter for those programs that have entered clinical development prior to the end of the collaboration term. For all other programs resulting from the collaboration, Gilead will make a $150 million opt-in payment per program and will owe no subsequent milestones. Galapagos will receive tiered royalties ranging from 20-24% on net sales of all products licensed by Gilead in all countries outside Europe as part of the agreement.Criticism of Gilead by Van de Stolpe. Perhaps Gilead, says Van de Stolpe, should have chosen a different strategy and only asked for approval for the high dose (200mg). Then the FDA would not have compared the two doses and judged the high dose on its own merits. Approval of only the high dose would have offered sufficient prospects to compete with similar drugs, precisely because they have so far only been allowed in the lower doses in the US. This criticism of Gilead's strategic choice, however cautiously formulated, is remarkable. The activities of Galapagos and Gilead have been closely intertwined since the two companies entered into an intensive partnership last year. Van de Stolpe would not be Van de Stolpe if he also could not spot a bright spot. In this case, it is the fact that Galapagos has now acquired all European property rights to filgotinib. It means that Gilead is leaving the marketing in Europe to Galapagos. 'That's a really great step. We can now make all the decisions ourselves, without ending up in a viscous consultation structure.'Van de Stolpe: I don't worry about the stock price. I've been in this world too long for that. The stock price is currently of less strategic importance. We have enough cash in hand if we want to make acquisitions, we don't have to issue shares for that. Van de Stolpe does notice some unrest among the employees. The next 2 years will be crucial, according to Van de Stolpe. Whether the Gilead/Galapagos deal proves inspirational in the future will depend on how many of Galapagos’s drug candidates actually succeed.
Ik denk eerder dat het commitment van Gilead over and out is wat betreft filgotinib. De meeste believers van de laatste jaren zijn hier opeens verdwenen van het forum. Was ook wel benieuwd naar de visie van oa Wic32 op heel deze situatie. Is filgotinib dan toch niet "best in class" en het veilgste van alle jaks zoals ons altijd voorgehouden werd? De FDA gaat dit toch niet uit hun duim zuigen. Onno beseft goed dat hij veel vertrouwen kwijt is bij de aandeelhouders maar wat zijn wij daar mee. Ik weet dat biotech risicovol is maar ik kan mij toch niet van de indruk ontdoen dat wij misleid zijn door Onno en niet alleen de kleine aandeelhouders maar ook Van Herk en Gilead.
