www.iex.nl/Forum/Topic/1320158/32/Beu... Dank flosz.
Morphosys Annual Report 2014 New mechanisms for treating inflammatory diseases such as rheumatoid arthritis, osteoporosis or osteoarthritis are being examined in cooperation with the Belgian company Galapagos NV with the aim of developing new antibody therapies to treat these diseases. The first candidate from this cooperation – MOR106 – entered preclinical development in the reporting year. Both companies contribute their core technologies and expertise as part of this alliance. In accordance with the agreement, Galapagos and MorphoSys share research and development costs and all future revenues equally. P.21 Future Research and Development The Company’s R&D budget for proprietary drug development will rise significantly in 2015 in comparison to previous years. The majority of these investments will flow to the clinical develop- ment of the most advanced drug candidates MOR208, MOR202 and MOR209. Further investments are planned in the areas of target validation and antibody development as well as in the area of tech- nology development. The steps planned for the Company’s proprietary portfolio in 2015 are expected to include: • Continuation of a phase 2 trials of MOR208 in NHL and B-ALL • Initiation of further combination trials for MOR208 in NHL • Continuation of a phase 1/2a trial of MOR202 with additional cohorts with weekly dosing as well as a combination of MOR202 with pomalidomide and lenalidomide • Initiation of the phase 1 trial for MOR209/ES414 in mCRPC as part of the cooperation with Emergent • Continuation of the co-development program for MOR106 with Galapagos • In-licensing of one or more target molecules or compounds for strengthening the proprietary development portfolio • Further development of the lanthipeptide technology • Initiation and continuation of new development programs in the area of antibody discovery and preclinical development P.47 In November 2008, MorphoSys and Galapagos announced the beginning of a long-term joint drug discovery and development cooperation. The goal of the cooperation is to explore novel mechanisms for the treatment of in- flammatory diseases and to develop antibody therapies against these dis- eases. The agreement covers all activities ranging from the probing of target molecules to the completion of clinical trials for novel therapeutic antibodies. Subsequent to the demonstration of clinical efficacy in hu- mans, the programs will be out-licensed to partners for further develop- ment, approval, and commercialization. Both companies provided their core technologies and expertise within the scope of the alliance. Along with the use of its adenovirus-based platform for the exploration of new target molecules for the development of antibodies, Galapagos provided access to already identified target molecules that are associated with bone and joint diseases. MorphoSys provided access to its antibody technolo- gies used for generating fully human antibodies directed against these target molecules. Under the terms of agreements, both Galapagos and MorphoSys bear the costs of research and development. In July 2014, the collaboration advanced into the preclinical development of MOR106, an antibody from MorphoSys’ next-generation antibody library Ylanthia di- rected against a novel Galapagos target molecule. The antibody will be co-developed in the area of inflammatory diseases. P.138www.morphosys.com/sites/default/files...

MorphoSys Presents First Safety, Pharmacokinetic and Efficacy Data for MOR202 in Multiple Myeloma at ASCO Meeting Clinical Activity and Long-lasting Tumor Control Already Observed at Low Doses MOR202 is safe and tolerable in weekly and biweekly doses MOR202 showed early signs of clinical activity with cases of long-lasting tumor control MOR202 demonstrated best-in-class infusion tolerability as consistent 2-hour infusion with no infusion reaction in combination with dexamethasone Overall, the first clinical MOR202 data hint towards a balanced and potentially best-in-class safety/efficacy profile Preclinical data demonstrate synergistic potential of MOR202 and pomalidomide, an established immunomodulatory agent in relapsed/refractory multiple myeloma therapy Dose escalation to 16 mg/kg weekly plus dexamethasone is ongoing; combination with pomalidomide and lenalidomide plus dexamethasone to start shortly MorphoSys AG (FSE: MOR; Prime Standard Segment, TecDAX; OTC: MPSYY) today published first safety and efficacy data on its proprietary drug candidate MOR202. The data are from a phase 1/2a study in 42 heavily pretreated patients with relapsed/refractory multiple myeloma. MOR202 is a fully human monoclonal HuCAL antibody targeting CD38, a highly expressed and validated target in multiple myeloma. The preliminary clinical data, which were presented today at the 2015 American Society of Clinical Oncology Annual Meeting (ASCO), show that MOR202 was safe and well tolerated with a best-in-class infusion tolerability as a 2-hour infusion. Infusion reactions in patients receiving MOR202 without dexamethasone were mostly mild to moderate and mainly limited to the first infusion. In patients receiving MOR202 in combination with dexamethasone, no infusion reactions were observed. MOR202 demonstrated promising early signs of activity, and cases of long-lasting tumor control were already observed in early cohorts. The ongoing phase 1/2a, open-label, multi-center, dose-escalation study is being conducted in several centers in Germany and Austria. The study is evaluating the safety and preliminary efficacy of MOR202 as monotherapy and in combination with pomalidomide and lenalidomide plus dexamethasone in patients with relapsed/refractory multiple myeloma. The primary endpoints of the trial are the safety, tolerability and recommended dose of MOR202 alone and in combination with immunomodulatory drugs (IMiDs). Secondary outcome measures are pharmacokinetics and preliminary efficacy based on overall response rate, duration of response, time-to-progression, and progression-free survival. The data presented today at the ASCO 2015 meeting show that MOR202 was safe and tolerable and could be administered as a 2-hour infusion. Infusion-related reactions occurred in 13 patients (37%) receiving MOR202 without dexamethasone, mainly limited to the first infusion. All infusion reactions were grade 1-2 except for one patient with grade 3. No infusion reactions occurred in patients who received dexamethasone. The maximum tolerated dose (MTD) has not been reached. The best response to date, a very good partial response (VGPR), was achieved in the 4.0 mg/kg weekly dose cohort plus dexamethasone. Additionally, one minimal response (MR) and ten cases of stable disease (SD) in other dose groups starting as low as 1.5 mg/kg biweekly were reported. The preliminary pharmacokinetic data indicate the potential for full target occupancy with a dose of 16 mg/kg administered weekly. The study is ongoing and the higher doses of MOR202 are being evaluated in combination with dexamethasone. Further cohorts in which patients will receive MOR202 up to 16 mg/kg weekly in combination with the IMiDs pomalidomide and lenalidomide plus dexamethasone will commence shortly. "MOR202 has shown clinical activity, which is already very promising. The safety and tolerability profile, especially regarding infusion reactions, opens up the clear potential to differentiate our compound from other anti-CD38 therapies currently in development," commented Dr. Arndt Schottelius, Chief Development Officer of MorphoSys AG. "We are pleased with this first snapshot of MOR202's clinical potential. The data will mature over the coming months and we will provide updates at future medical conferences." MorphoSys additionally presented today promising preclinical data demonstrating the synergistic potential of MOR202 and pomalidomide, an established immunomodulatory agent in relapsed/refractory multiple myeloma therapy. This synergistic benefit was between 1.2-fold and 3.1-fold above the theoretical benefit of combining the drugs and resulted from multiple mechanisms, namely direct cytotoxicity, CD38 up-regulation and activation of effector cells. Dr. Schottelius added: "We believe MOR202 is a very attractive combination partner in multiple myeloma. The data presented today support the rationale of combining MOR202 with established multiple myeloma drugs such as lenalidomide and pomalidomide in cohorts which will commence shortly in the ongoing study."

April 07, 2016 / 8:30 am, CEST MorphoSys AG (FSE: MOR; Prime Standard Segment, TecDAX; OTC: MPSYY) and Galapagos NV (Euronext & NASDAQ: GLPG) announced today that dosing of MOR106, a first-in-class human monoclonal antibody, in healthy volunteers has been initiated in a phase 1 study. MOR106 was jointly discovered by MorphoSys and Galapagos under their collaboration, and has a novel mode of action with potential application in inflammation. The primary objective of the phase 1 study is to evaluate the safety and tolerability of single doses of MOR106. The study is randomized, double-blind, placebo-controlled and conducted in a single center in at least 56 healthy volunteers in Belgium, evaluating single ascending doses (SAD) administered as intravenous infusion. This phase 1 study is characterized by its adaptive design, which enables the initiation of a subsequent multiple ascending dose (MAD) study in patients, depending on the outcome of the SAD study in healthy volunteers. The study's secondary objective is to characterize the pharmacokinetic profile of MOR106 as well as monitor the occurrence of anti-drug antibodies as a measure of immunogenicity with MOR106. Topline results of the complete study, including the potential subsequent MAD part in patients, are expected in the second half of 2017. "We are delighted to see the first antibody program from our alliance with Galapagos entering the clinical development stage. MOR106 is MorphoSys's fifth proprietary antibody program in clinical development and the first from our novel Ylanthia technology platform. We are excited about the growing value and maturity of our development pipeline. The start of this very innovative clinical development program also reflects the high value of our ongoing collaboration with Galapagos", commented Dr. Arndt Schottelius, Chief Development Officer of MorphoSys AG. "The alliance with MorphoSys has produced a first-in-class candidate human monoclonal antibody," said Piet Wigerinck, Chief Scientific Officer of Galapagos. "MOR106 is the 10th Galapagos candidate with a new mode of action to be brought into the clinic." About MOR106 and the antibody collaboration MOR106 is a human IgG1 monoclonal antibody for treatment of inflammatory diseases. MOR106 arises from the alliance initiated by Galapagos and MorphoSys in 2008, in which both companies contribute their core technologies and expertise. Galapagos provides the disease-related biology including cellular assays and targets discovered using its target discovery platform. MorphoSys contributes its Ylanthia antibody technology to generate fully human antibodies directed against the target and contributes full CMC development of this compound. Galapagos and MorphoSys will continue to equally share the research and development costs, as well as all future revenues.www.morphosys.com/media-investors/med...
