Zoek de verschillen: Trial 1:clinicaltrials.gov/ct2/show/NCT003711... First Received: August 31, 2006 Adenovirus Vaccine for Malaria This study is ongoing, but not recruiting participants. ponsor: National Institute of Allergy and Infectious Diseases (NIAID) roup A: 10^8 vp/mL (15 persons) or placebo (3 persons) Group B: 10^9 vp/mL (15 persons) or placebo (3 persons) Group C: 10^10 vp/mL (15 persons) or placebo (3 persons) Group D: 10^11 vp/mL (15 persons) or placebo (3 persons) Locations United States, California Stanford University Stanford, California, United States, 94305 United States, Tennessee Vanderbilt University Nashville, Tennessee, United States, 37232 Trial 2: First Received: November 19, 2009clinicaltrials.gov/ct2/show/NCT010184... Adenovirus Type 35 Based Circumsporozoite Malaria Vaccine in Burkina Faso This study is not yet open for participant recruitment. Sponsor: National Institute of Allergy and Infectious Diseases (NIAID) Group A: 10^9 vp/mL (10 persons) or placebo (3 persons) Group B: 10^10 vp/mL (10 persons) or placebo (3 persons) Group C: 5*10^10 vp/mL (10 persons) or placebo (3 persons) Group D: 10^11 vp/mL (10 persons) or placebo (3 persons) Locations Burkina Faso Centre National de Recherche et de Formation sur le Paludisme West Africa, Burkina Faso Dus: Eerst in de VS testen voor veiligheid, daarna mag je in ontwikkelingslanden trials houden (ging met tbc vaccin net zo). Dus nu komt de eerste phase I malaria trial op Afrikaanse bodem eraan. Minieme wijzigingen t.o.v. US trial: 1) 10 i.p.v. 15 proefpersonen per groep. Edit: in US de drie vaccinaties per persoon verspreid over 6 maanden, in Burkina Faso verspreid over 3 maanden. 4 groepen x 6 maanden = 24 maanden. 4 groepen x 3 maanden = 12 maanden.

Babs

Heerlijk.
Parasites In Cambodia Becoming Resistant To Antimalaria Drug Last update: 2/25/2010 4:46:01 AM LONDON (Dow Jones)--Parasites are developing resistance to artimisinin, one of the most important antimalaria drugs, the BBC reports on its Web site Thursday, citing experts from the Malaria Consortium. Artimisinin has been highly effective, particularly in places where resistance to other drugs has developed, but now some patients along Cambodia's border with Thailand are taking longer to respond to the treatment, the report says. (Euronext, NASDAQ: CRXL; Swiss Exchange: CRX), today announced that GlaxoSmithKline (GSK) signed a license agreement allowing GSK to research a recombinant protein on the PER.C6® platform. Financial terms of the agreement were not disclosed. "We are very pleased to enter into this collaboration with GSK", said Ronald Brus, Crucell's Chief Executive Officer. "This agreement further underscores the potential of our PER.C6® production technology for generating recombinant proteins." "The PER.C6® production technology continues to surpass our expectations as a robust manufacturing platform with its ever extending and global reach into the Research and Development departments of major healthcare players," adds Karen King, President of DSM Biologics. "This agreement with GSK brings us closer to our mission of making PER.C6® technology a gold standard for industry use.") Pubmed-draad. Bv: www.iex.nl/forum/topic.asp?forum=228&... 3. Expected news flow • Partnership for malaria vaccine development On the last page, the expected news flow for the year, first of all, a partnership for a malaria vaccine development. As Cees just mentioned, we have successfully completed our clinical trial in the United States, and we feel that with the strong technology base that we have for a T-cell malaria vaccine, it is a prime candidate for partnering and could actually grow to one of the most promising malaria vaccines in the world. With respect, I would just like to come back on the malaria. I think if you look into what is currently going on in malaria, we see that GlaxoSmithKline is doing tremendously on a vaccine that they currently are developing in Africa. And I think basically what that demonstrates, it is doable to come up with a malaria vaccine, because the T-cell responses and the clinical responses surely suggest that it is much better than placebo. However, looking at the preventive rates that we observe, it's clear that it needs to be improved, that a 40% efficacy is clearly not good enough on the longer run. And I think you've seen public statements of [Andrew Witty] on this subject. You know that there's a lot of movement in this arena; hence, our Company would like to position ourselves as being at the forefront of what is going to happen in malaria. And therefore, we KNOW that we NEED ONE DAY TO JOIN FORCES WITH THE OTHER LEADER IN THIS PACK! And that's why we're positioning ourselves, and that's what we're trying to achieve. Let me answer the question on malaria. You've seen the positive results that we had in Phase I and the new collaboration that we announced. Forming a partnership, we would do to accelerate time to market or obtain better access to markets. So it might mean that we create combinations with other parties' products to get even better products than the ones we have in the clinic. I think, Brigitte, you will have to wait until we come with the announcement so we can further clarify why we would have formed such partnerships. (M.a.w.: we zijn in gesprek).www.huginonline.nl/CRXL/files/2010021...
Crucell Announces Collaboration with GlaxoSmithKline Biologicals on Second Generation Malaria Vaccine Candidate Leiden, the Netherlands (April 6, 2010) - Dutch biopharmaceutical company Crucell N.V. (NYSE Euronext, NASDAQ: CRXL; Swiss Exchange: CRX) today announced that it has signed a binding letter of agreement with GlaxoSmithKline Biologicals (GSK) to collaborate on developing a second generation malaria vaccine candidate. Pre-clinical data from earlier studies indicated significantly enhanced immune responses against the malaria parasite (circumsporozoite stage of the Plasmodium falciparum) when Crucell's Adenovirus (AdVac®) technology and GSK's RTS,S/AS technology are used in combination, versus either component alone. Under the terms of the letter of agreement, Crucell will contribute its recombinant malaria vaccine candidate, Ad35-CS, based on Crucell's AdVac® technology and PER.C6® manufacturing platform and GSK will contribute its late stage malaria vaccine candidate RTS,S/AS. Financial details of the agreement were not disclosed. In October 2003 Crucell announced a program with the Walter Reed Army Institute of Research and GSK. The parties entered into a Cooperative Research and Development Agreement (CRADA) to evaluate Crucell's malaria vaccine candidate in pre-clinical studies. Crucell's vaccine candidate was tested as a stand-alone and in combination with GSK's malaria vaccine candidate RTS,S/AS. The tests conducted were designed to assess whether a combination of the GSK vaccine candidate with Crucell's vaccine candidate could lead to improved results. These pre-clinical studies showed significant enhancement of the immunological response when both vaccines were used in combination[1]. This collaboration now intends to progress this concept into human clinical studies. The companies will seek third party funding to advance the Ad35-CS and RTS,S/AS prime-boost candidate into a Phase I/IIa clinical challenge trial in the United States. Pending the results of the Phase I/IIa trial, the companies expect to advance the prime boost candidate in further clinical studies with the support of public or non-profit partners who are interested in accelerating the development of a malaria vaccine. "We are very excited about this collaboration with GSK, which brings new impulse to the potential of eradicating malaria as one of the top three killers in the world," said Ronald Brus, Chief Executive Officer at Crucell. "Malaria currently represents one of the most prevalent infections in tropical and subtropical areas, causing close to 900,000 deaths every year, mostly amongst children. I am confident a partnership of this kind will allow us to take a vital step towards our goal of bringing meaningful innovation to global health." ********************** Malaria Finally, to eradicate the disease, we will almost certainly need a malaria vaccine, which is the highest-risk malaria work we fund. The key here is that researchers are pursuing a lot of different ideas, so that if one fails, there are still several other options. One partially effective vaccine candidate, known as RTS,S, has started its Phase III trial, which is an important step. Other vaccine approaches are at an earlier stage and they also look very promising. Scientists are combining some of these other vaccine efforts with RTS,S to raise its effectiveness and duration, an approach that could lead to a highly efficacious vaccine in 8 to 15 years. www.gatesfoundation.org/annual-letter... ********************* Developing new malaria vaccine Professor Moriya Tsuji tells International Innovation about the efforts to ‘wipe malaria off the face of the earth’ The most advanced malaria vaccine candidate so far is called RTS,S vaccine, which consists of a fusion protein of a portion of the circumsporozoite (CS) protein with the hepatitis B surface antigen, incorporated in a mixture of three adjuvants. This vaccine is now in Phase III trial. However, the effi cacy of this vaccine as of Phase II trials has been around 50-60 per cent against disease, and the protective immunity seems to be primarily mediated by humoral response. Therefore, for the purpose of improving the effi cacy of malaria vaccine, we are aiming to develop a new malaria vaccine that can induce; 1) cellmediated immunity (CMI) against 2) novel malarial antigen(s). Furthermore, has your organisation collaborated with international partners – has the collaborative approach worked? In the past, my group had collaborated with the Crucell Holland B.V, and together, we found that adenovirus serotype 35 (Ad35) can be a potent vaccine vector as a malaria vaccine. This study became a basis for the Crucell to move onto some of the currently ongoing clinical trials using Ad35 vector for a malaria vaccine. www.adarc.org/files/Tsuji_Malaria_vac... Malaria: The end of the beginning Hill's doubts about the vaccine bolster a more general frustration with what he sees as GSK's go-it-alone approach, “unconnected to the other 12 or 15 groups developing vaccines” (see table 1). He and others want different vaccines to be combined with RTS,S in Phase II testing, suspecting efficacy might be greatly enhanced. Heppner, for example, says that results of studies he and his colleagues have carried out on macaques indicate that combining an adenovirus-based vaccine made by the Netherlands biotech company CruCell with RTS,S would offer much better effects[10]: “My hope is that a way can be found to evaluate this clinically just as we've done for earlier improvements of RTS,S.” Ballou says that although progress in studying this combination has been stalled for “various business reasons”, several collaborative efforts continue.
